摘要
利用Biotin-16-dUTP标记的cDNA探针分子杂交检测分析了新疆梨树上的3种主要病毒(梨环纹花叶病毒、梨脉黄病毒、苹果茎沟病毒)并优化了杂交反应体系。结果表明,这3种病毒探针的检测灵敏度分别为5、10和10 pg。用文中的方法提取的RNA均能在10-3稀释后显色。该方法与RT-PCR方法对样品带毒状况的检测结果一致。
This paper used cDNA probe molecular hybridization assay to detect three economically important viruses on Xinjiang's commercial pear cultivars: Pear vein yellows virus (Foveavirus), Pear ring pattern mosaic virus(Trichovirus), Apple stem grooving virus (Capilloviru), and to optimize hybridization reaction system. The results indicated cDNA probe coloration sensitivity of these three symptomless viruses are 5 pg, 10 pg and 10 pg differently. Total RNA isolated by this assay can coloration after diluted in 10-3 times. We gained identically detection results with RT-PCR assays to several samples.
