实时荧光定量PCR检测端粒酶逆转录酶mRNA在急性白血病中的表达

摘要 目的研究人类端粒酶转录酶(hTERT)在急性白血病的表达和意义。方法应用实时定量PCR(Real-time PCR)技术检测30例急性白血病骨髓单个核细胞及25例正常人骨髓单个核细胞中hTERT mRNA的表达水平,分析其表达水平与急性白血病恶性程度的关系。结果hTERT在急性白血病细胞中的中位表达量为0.00244(0~0.0320),明显高于正常人骨髓单个援细胞中hTERT的中位表达量2.79×10^-4(0~0.0078)(P〈0.001)。但在ANLL和ALL细胞中对比表达无明显差异(P=0.742)。结论在急性白血病中hTERT明显高于正常对照,hTERT是急性白血病诊断的特异性指标之一。 Objective To detect the expression of human telomerase reverse transcriptase (hTERT) in acute leukemia and its significance, Methods Quantitative real-time PCR methods were used to examine the expression of hTERT mRNA in hone marrow mononuclear cells (MNCs) from 30 acute leukemia patients and 25 controls, then the correlation between the expression of hTERT mRNA and the malignant nature of acute leukemia was analyzed. Results hTERT mRNA expression in acute leukemia cells (0.00244, 0~0.320) was significantly higher than that in normal cells (0.000279, 0~0.0078) (P 〈 0.001). Its expression had no significant difference between ANLL and ALL cells. Conclusions hTERT level increases in acute leukemia, which indicates that hTERT is one of the special markers for the diagnosis of acute leukemia.
