摘要
目的探讨催乳素(PRL)对B淋巴细胞生长的调节作用。方法将生理浓度(10 ng.ml-1)的PRL加到IM-9细胞中培养72 h,以MTT比色分析法检测细胞增殖情况。结果空白对照组、脂多糖(LPS)组、PRL组、LPS+PRL组、LPS+溴隐停(Br)组、LPS+PRL+Br组A值分别为1.00±0.09、1.11±0.14、1.21±0.23、1.38±0.44、0.95±0.14、0.89±0.11。与空白对照组比较,PRL(t=2.689,P<0.01)、LPS(t=2.292,P<0.05)均能促进IM-9细胞的增殖;与LPS组相比,加入Br后对IM-9细胞的增殖有抑制作用(t=2.556,P<0.05);与LPS-PRL组相比,加入Br后对IM-9细胞的增殖亦有抑制作用(t=3.417,P<0.01)。结论人PRL能明显地促进IM-9细胞的增殖,Br对IM-9细胞的增殖有抑制作用。
Objective To explore the regulatory effects of prolactin (PRL) on proliferation of human B lymphocyte. Methods The addition of PRL( 10 ng·ml^-1) , physiological concentration, into IM-9 cell lines for cultttring for 72 h was to detect cellular proliferation using MTT colorimetric analysis. Results The absorption value of the blank control group, Lipopolysaccaride (LPS) group, PRL group, PRL plus LPS group, LPS plus Br group, LPS plus PRL plus Bromoeripfine (Br) were 1.00 ± 0.09,1.11 ± 0.14,1.21 ± 0.23,1.38 ± 0.44, 0, 95 ± 0.14 and 0.89 ± 0.11, respectively. Compared with the blank oontrol group, PRL group ( t = 2. 689, P 〈 0.01 ) and LPS group ( t = 2. 292, P 〈 0.05) stimulated the proliferation of IM-9 cell lines. Compared with LPS group, the addition of Br inhibited the proliferation of IM-9 cell lines( t = 2.556,P〈0. 05), Compared with the LPS plus PRL group, the addition of Br also inhibited the proliferation of IM-9 cell lines ( t = 3. 417, P〈 0.01 ). Conclusion Human PRL promotes obviously the proliferation of IM-9 cell line, however, Br plays a role in inhibition of proliferation of IM-9 cell lines.
