正常大鼠角膜NF-κB及其下游因子ICAM-1、VEGF的表达与意义 被引量:1

摘要 目的:研究核因子-κB(NF-κB)及其下游因子细胞间黏附分子-1(ICAM-1)、血管内皮生长因子(VEGF)在正常角膜中的表达规律,为进一步研究角膜移植排斥反应的发病机制及治疗提供新的思路。方法:健康清洁级Wistar大鼠5只,体重200~250g。10%水合氯醛溶液腹腔注射麻醉后立即摘除眼球共10个,剪下完整角膜(带角膜缘),置于4%多聚甲醛溶液固定,常规石蜡包埋,切片。每份标本连续制作4张4μm切片,1张行HE染色,其余3张分别用于NF-κB、ICAM-1、VEGF的免疫组织化学染色(SABC法)检测。结果:正常的大鼠角膜中,NF-κB及其下游因子ICAM-1、VEGF在角膜上皮层细胞内有较弱的基础表达,主要表达在基底层细胞;而在角膜基质层及内皮细胞未见阳性表达;NF-κB、ICAM-1、VEGF在正常大鼠角膜的表达部位及表达强度具有良好的一致性。结论:NF-κB可能通过调控ICAM-1、VEGF等基因的表达,参与角膜移植排斥反应及角膜新生血管形成,起着中心调控者的作用。 Objective To investigate the expressions of nuclear factor-κB(NF-κB), intercellular adhesion molecule-1 (ICAM-1) and vascular endothelial growth factor (VEGF) in rat corneas, and to analyze their roles in the corneal allograft rejection. Method Five Wistar ratsrwere applied, and the expressions and distributions of NF-κB, ICAM-1 and VEGF in cornea were detected with immunohistoehemical methods and image analysis. Results NF-κB and ICAM-1, VEGF were weekly expressed in basal layer of corneal epithelium in rat cornea, and not expressed in keratocytes and corneal endothelial cells. The location of expression of NF-κB was similar to ICAM-1 and VEGF, and had nicely correlations. Conclusion NF-κB may regulate the expression of genes associated with graft rejective and corneal neovascularization (CNV), such as ICAM-1 and VEGF expression, and thus play a role of center-control in the occurrence and development of corneal graft rejective reaction.
