摘要 目的建立HPLC-ELSD法测定芪枣颗粒中黄芪甲苷含量的方法。方法色谱条件为:Shim-packC18色谱柱(250mm×4.6mm,5μm);ELSD检测器,漂移管温度:40℃;雾化气:氮气;载气压力:3.5bar;柱温:30℃;流动相:乙腈-水(30∶70);流速:1mL/min。结果黄芪甲苷在2.51~50.2μg范围内呈线性关系,r=0.9998;加样回收率为98.13%,RSD=1.28%。结论该方法灵敏,准确,可用于芪枣颗粒的质量控制。 Objective To determine the content of astragaloside IV in Qi Zao Granules. Methods Equipped with the evaporative light scattering detector (ELSD), a RP - HPLC method was established. The chromatographic column was C 18 and the column temperature was at 30℃. The drift tube temperature was at 40 ℃ and the evaporative gas was nitrogen with pressure of 3.5 bar. The mobile phase was acetonitrile - water(30 : 70) and the flow rate was lmL/min. Results This method showed a good linear relationship in the range of 2.51 - 50.2 μg astragaloside IV. The average recovery was 98.13 % with RSD being 1.28 % . Conclusion The method is sensitive and accurate and can be used for the quality control of Qi Zao Granules.
