脂可平对实验性早期动脉粥样硬化氧自由基及主动脉ICAM-1表达的影响 被引量:10

摘要 目的:研究脂可平对大鼠早期动脉粥样硬化氧自由基及细胞间黏附分子(ICAM-1)表达的影响。方法:应用高脂饲料复制SD大鼠早期动脉粥样硬化模型,分别给以脂可平、辛伐他汀片混悬液灌胃,实验10周后,全自动生化分析仪检测各组大鼠血脂、血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,用免疫组化和RT-PCR方法检测主动脉壁ICAM-1及其mRNA的表达水平;苏木素伊红(HE)染色观察主动脉组织形态学变化。结果:两治疗组血脂、血清MDA含量均明显降低,SOD活性显著升高,不同程度的改善了主动脉组织病理损伤,ICAM-1及其mRNA的表达水平显著下降。结论:脂可平抗动脉粥样硬化作用的机制可能与其抗氧化、抗粘附等保护血管内皮细胞功能密切相关。 Objective: To investigate the effects of Zhikepingon the oxyradical andintercellular adhesion molecule-1 (ICAM-1) of experimental early atherosclerosis. Method: The model of SD rat early atherosclemsis was induced by cholesterol diet. The suspension of Zhikepingand simvastatin were administered intragastrically, respectively. After 10 weeks, the serum lipids, supemxide dismutase (SOD) and maleic dialdehyde (MDA) were detected by automatic biochemistry analyzer. ICAM-1 and its expression of mRNA in aortic wall were detectedby immunohistochemistryand RT-PCR, respectively. Aortic histomorphology was cbserved by HE stainning. Result: The results showed that the serum lipids, supemxide dismutase (SOD) and maleic dialdehyde (MDA) of treated groups were obviously improved as compared with those of the controlgroup. The tissue pathologicaldamage was improved in different degree,and ICAM-1 and its expression of mRNA was de creased obviously. Conclusion: It is suggested the mechanism of anti-atherosclerosis of Zhikeping have close relationship with the function of its anti-oxidizing and anti-adhesiveness that can protect aortic endothelial cell.
