决明子中丝氨酸蛋白酶抑制剂的分析

Screening Serine Protease Inhibitor from Seed of Cassia obtusifolia by Gelatin-Polyacrylamide Gel

目的确定决明子中丝氨酸蛋白酶抑制剂的种类和数量。方法材料经过明胶-PAGE和明胶-SDS-PAGE后,分别用胰蛋白酶进行酶解,考马斯亮兰染色。结果电泳胶板经胰凝乳蛋白酶处理后,观察到一条清晰的蛋白染色带。结论决明子中含有一种胰凝乳蛋白酶抑制剂,该抑制剂有较强的抗还原能力,但在强酸环境中易发生变性。

Objective To determine the species and quantities of serine protease inhibitors in Jue-ming-zi（ Cassia obtusifolia L）. Methods After gelatin-PAGE and gelatin-SDS-PAGE,the gel was incubated in Tris-HCl buffer, containing trypsin or chymotypsin,and then was stained with Coomassie blue 11250. Results A clear inhibitor band on the gel was observed after chymotrypsin treatment. Conclusion It was found that the existence of a chymotrypsin in Jue-ming-zi, which had resistance to reducing agents and tendency to denaturation under strong acidenvironments.
