摘要 目的:研究c-myc小干扰RNA对白血病细胞凋亡的影响,探寻白血病基因治疗的新方法。方法:应用针对c-myc的小干扰RNA转染HL-60细胞48 h后,倒置显微镜下观察细胞形态学变化,琼脂糖凝胶电泳法检测DNA梯形带和流式细胞仪Annexin V-FITC/PI法检测细胞凋亡。结果:c-myc小干扰RNA作用后,HL-60细胞生长受抑,可见细胞核固缩,胞桨内出现凋亡小体。阴性对照组和空白对照组没有明显变化。DNA电泳干扰组出现典型的梯状条形带,对照组未出现明显凋亡梯形带。流式细胞仪分析干扰组细胞凋亡率可达26.21±0.75%,明显高于阴性对照组的4.58±0.48%和空白对照组的3.76±0.30%。结论:c-myc小干扰RNA对HL-60细胞有明显的抑制作用,可抑制细胞增殖,导致细胞凋亡,有望成为白血病靶向基因治疗的新工具。 Objective: To study the effect of the c-myc small interference RNA on apoptosis of leukemic cells. Methods : After c-myc siRNA was transfected into HL-60 cells for 48 hours , growth apperance and morphology of the cells were observed with microscope , agarose gel electrophoresis was performed for the DNA ladder and the percentage of apoptosis cells was analyzed by flow cytometric annexin VFITC/PI. Results : After being acted by c-myc siRNA, the growth of HL-60 cells was inhibited , the nuclei of the cells shrinked and the apoptosis body was observed in the plasma of the cells. The DNA ladder was clearly detected in agarose gel electrophoresis , FCM analysis showed that the percentage of apoptosis cells in c-myc siRNA group was 26.21 ± 0.75 %, significantly higher than that in control siRNA group 4.58 ± 0.48 % and non-transfection group3.76 ± 0.30%. Conclusion : c-myc siRNA could significently inhibit HL-60 cells , it inhibits HL-60 cells to proliferate and causes the cells to apoptosis, c-myc siRNA may become one of the new tools of targeting gene therapy to leukemia.
