摘要
目的探讨HCMV感染对体外培养人血管内皮细胞凋亡的影响。方法建立HCMV-AD169株感染体外培养人脐静脉血管内皮细胞模型,应用AV/PI双染法检测HCMV感染后ECs的凋亡变化。结果体外情况下HCMV感染后不同时相点(12、24、48、72h),ECs的凋亡率分别为(10.6±4.6)%,(23.8±6.9)%,(39.4±8.9)%,(31.4±7.8)%,明显高于对照组细胞。结论HCMV感染诱导ECs凋亡的异常改变可能参与了动脉粥样硬化的发生发展。
Objective To study the effects of human cytomegalovirus infection on the apoptosis of cultured endothelial cells. Methods Building the model of human cytomegalovirus infect human endothelial cells in vitro, the apoptosis of ECs was detected by flow cytometry of annexin V-FITC/PI method. Results At different infection time( 12h, 24h,48h, 72h), the apoptosis rates were (10.6±4.6) %, (23.8±6.9) %, (39.4±8.9) % and (31.4±7.8) % respectively, and those were signifieantly higher than that of control group. Conclusion HCMV infection could develop the apoptosis of endothelial cells in vitro, and this may play an important role in the pathogenesis of atherosclerosis.
