摘要
目的:验证大戟科多年生肉质草本植物猫儿眼多糖对力竭运动小鼠心肌线粒体的保护作用。方法:①实验于2004-11/12在甘肃政法学院公安分院实验中心完成。选用出生80d的雄性昆明种小鼠50只。随机将小鼠分为5组(每组10只):游泳+猫儿眼多糖低、中、高剂量组[按1.0,5.0,10.0g/(kg·d)剂量灌胃猫儿眼多糖;然后进行力竭游泳实验,沉入水下停留10s为止,1次/d],单纯游泳组(力竭游泳方法同给药组,灌胃等量蒸馏水),对照组(正常饲养,不游泳,灌胃等量蒸馏水)。连续干预7d。②给药7d后,心肌线粒体蛋白定量测定采用Bradford方法。心肌线粒体过氧化脂质、超氧化物歧化酶、谷胱甘肽过氧化物酶含量测定采用相应试剂盒。游离钙的测定参照Mccormark方法进行。用回归方程计算样品中肝糖原含量。③计量资料差异比较采用Dunntt单因素方差分析进行统计学处理。结果:小鼠50只均进入结果分析。游泳+猫儿眼多糖各剂量组小鼠连续给药7d,小鼠心肌线粒体丙二醛含量比单纯游泳组分别降低了13.5%,38.8%和59.9%(P<0.05~0.01);而超氧化物歧化酶含量比单纯游泳组升高了14.9%,55.8%和92.3%(P<0.05~0.01);谷胱甘肽过氧化物酶比单纯游泳组升高了72.2%,118.5%和174%(P<0.05~0.01);游离钙比单纯游泳组升高了11.8%,25.3%和56.0%(P<0.05~0.01);肝糖原含量比单纯游泳组增加了76.9%,101.9%和124.7%(P<0.01)。结论:猫儿眼多糖具有降低过氧化脂质的生成、提高力竭游泳小鼠心肌线粒体超氧化物歧化酶、谷胱甘肽过氧化物酶活性和肝糖原含量的功能,从而清除自由基,起到保护线粒体、抗疲劳作用。
AIM: To study protective effect of mooer yon polysaecharide on myocardial mitochondria of mice with exhaustive exercise.
METHODS: ① The experimetn was completed at the Experimental Center of Public Security, Gansu Institute of Political Science and Law from November to December 2004. Totally 50 male Kunming mice of 80- day old were selected and divided into 5 groups with 10 in each group. Mice were perfused with maoer yon polysaccharide according to various dosages [1.0, 5.0 and 10.0 g/(kg·d)] in swimming + low, middle and high dosage of maoer yon polysaccharide group. Then mice received exhaustive swimming test for 10 s underwater, once a day. Swimming group: Exhaustive swimming method was as the same as that in medication group, and mice were perfused with the same volume of distilled water. Control group: Mice were fed normally without swimming and perfusion. Intervention was kept for 7 days. ② Myocardial mitochondrial protein was measured with Bradford method 7 days after medication; levels of lipid oxidation, superoxide dismutase (SOD) and glutathione peroxidase (GSHPx) were measured with the corresponding kit; free Ca^2+ was determined on the basis of Mccormark method; and content of glycogen of liver was calculated with regression equation. ③Measurement data were compared with Dunntt single factor analysis of variance.
RESULTS: Totally 50 mice entered the final analysis. Contents of malondialdehyde (MDA) in myocardial mitochondria of mice in swimming + various dosages of maoer yon polysaccharide groups medicated for 7 successive days were decreased by 13.5%, 38.8% and 59.9%, respectively as compared with those in swimming group (P 〈 0.05-0.01); SOD contents were increased by 14.9%, 55.8% and 91.3%, respectively (P 〈 0.05-0.01); GSH-Px was increased by 72.2%, 118.5% and 174%, respectively (P 〈 0.05-0.01); free Ca^2+ was increased by 11.8%, 25.3% and 56.0%, respectively (P 〈 0.05-0.01); and glycogen of liver was increased by 76.9%, 101.9% and 124.7%, respectively (P 〈 0.01).
CONCLUSION: Maoer yon polysaccharide can decrease generation of lipid oxidation, increase contents of SOD, activity of GSH-Px, and content of glycogen of liver in myocardial mitochondria of mice with exhaustive exercise so as to clear free radical and to protect mitochondria and inhibit fatigue.
