摘要 目的:建立高效液相色谱法测定血浆中法舒地尔的浓度。方法:色谱柱:Hyperail C18(4.6mm×250 mm,10μm);流动相:0.05 mol·L-1磷酸二氢钾(pH 5.0)-甲醇(75:25);流速:1.0 ml·min-1;柱温:40℃;检测波长:276 nm。结果:法舒地尔在10-1000 ng·ml-1浓度范围内线性关系良好(r=0.999 1);低、中、高三种浓度相对回收率分别为89.73%,103.75%, 104.57%:日内、日间RSD均小于10%。结论:本法可行。 Objective: To establish an HPLC method for the determination of fasudil in blood plasma. Method: the Hypersil C18 column (4.6 mm× 250 mm, 10μm) was used. The mobile phase was 0.05 mol· L^-1 potassium dihydrogen phosphate-methanol (75 : 25). The flow rate was 1.0 ml· min^-1.The column temperature at 40 ℃ the Detection wavelength was at 276 nm. Result: The linear range of fasudil was 10 - 1000μg·L^-1 (r =0. 999 1 ) ,The recoveries were 84.86% ,101.51% ,104.57% in the low,middle and high concentrations, respectively and RSD were less than 10%. Conclusion: The method can be used for the determination the content of fasudil in blood plasma.
