基质金属蛋白酶-2和基质金属蛋白酶-9在非小细胞肺癌组织中的表达及临床意义

Expression of MMP-2 and MMP-9 in patients with primary non-small cell lung cancer (NSCLC) and its clinical significance

目的基质金属蛋白酶(matrixmetalloproteinase,MMP)-2和MMP-9在非小细胞肺癌(nonsmallcelllungcancer,NSCLC)组织中的表达及临床意义。方法用免疫组织化学方法和Westernblotting检测70例非小细胞肺癌组织中MMP-2和MMP-9蛋白的表达,并进行图像分析测定电泳条带的吸光度A值。结果70例非小细胞肺癌和30例正常肺组织中MMP-2、MMP-9表达阳性的例数分别为65.7%(46/70)、78.6%(55/70)和16.7%(5/30),23.3%(7/30)差别具有显著性。Ⅲ期肺癌MMP-2、MMP-9的平均吸光度A值明显高于Ⅰ、Ⅱ期。有淋巴结转移癌的MMP-2、MMP-9平均吸光度A值明显高于无淋巴结转移的病例,差异具有显著性(P<0.05)。肺腺癌中MMP-2、MMP-9平均吸光度A值高于肺鳞癌,差异具有显著性(P<0.05)。结论MMP-2、MMP-9在非小细胞肺癌组织中的表达增多,可能参与了非小细胞肺癌浸润和转移。

[Objective] To explore the expression of MMP-2, MMP-9 in patients with primary non-small cell hmg cancer （NSCLC）. [Methods] The expression of MMP-2, MMP-9 protein was detected in 70 NSCLC tissues by innunochenistry, and Western blotting followed by image analysis in order to detect the A value of expression. [Results] The positive rate of MMP-2, MMP-9 protein expression in NSCLC tissues was 65.7%（46/70）and 78.6% （55/70） respectively, which is higher than that in normal lung tissue. The A value of MMP-2 , MMP-9 expression in NSCLC tissues of Ⅲ stage was significantly higher than that in NSCLC tissues of Ⅰ , Ⅲ stages （P 〈0.05）. The A value of MMP-2, MMP-9 expression in NSCLC tissues of N1,2,3 groups was significantly higher than that in NSCLC tissues of NO group （P 〈0.05）. The expression of MMP-2, MMP-9 in NSCLC tissues of adenocarcinoma was significandy higher than that in NSCLC tissues of squamous cell earciuoma. [Conclusions] MMP-2, MMP-9 expression was related to lymph node metastasis and TNM stages of lung cancer. MMP-2, MMP-9 protein expression may be associated with the invasion and progression of NSCLC.
