摘要
目的检测RTECs在IL-18刺激下分泌细胞外基质(extracellularmatrix,ECM)纤维连接蛋白(fibronection,FN)、Ⅰ型胶原的量,为揭示肾间质纤维化机制提供新的途径。方法用分别含0、1、10和100ng/mL浓度的IL-18培养液于24孔板中体外培养细胞株HK-2,24、48和72h分别收集一次每一浓度上清液6孔,利用ELISA法分别检测上清液中FN、Ⅰ型胶原的含量。结果酶联免疫吸附法(ELISA)检测结果显示,RTECs上清液中FN、Ⅰ型胶原的含量存在IL-18浓度依赖关系,在分别作用24、48和72h时,不同浓度之间差异有显著性(P<0.0001)。结论IL-18通过刺激RTECs分泌ECM,可能在肾间质纤维化发挥着重要作用。
[Objective] To reveal the new mechanism of renal interstitial fibrosis by analyzing the fibroncction (FN) and Ⅰtype collagen( ⅠCOL) in supematant fluid of RTECs stimulated by IL-18. [Method] The extraceHular matrixes, FN and ⅠCOL, in supernatant fluid of RTECs were quantitated by enzyme-linked immunosorbent after RTECs stimulated by different concentration and time of IL-18. [Results] The quantity of FN and I COL in supernatant fluid was related to concentration and stimulating time of IL-18. The secretory volume of them was increasing along with concentration of IL-18 and the difference of different concentration groups,when stimulated in 24, 48 and 72 h respectively, were significant (P 〈0.0001). [Conclusion] IL-18 can increase the exudation of extracellular matrixes in RTECs, which perhaps plays important role in renal interstitial fibrosis.
