PI3K/AKT特异性抑制剂LY294002对胆管癌细胞化疗增敏作用的研究 被引量:4

摘要 目的探讨磷脂酰肌醇-3-激酶抑制剂(P I3K)LY 294002[22(42吗啉基)282苯基24氢212苯并吡喃242酮]对胆管癌化疗效果的影响。方法将LY 294002用于胆管癌细胞系QBC 939,M TT法检测单独使用5-Fu、MM C、塞莱西布及联合磷酸化抑制剂LY 294002,对体外培养的胆管癌细胞系QBC 939的抑制作用;流式细胞技术检测QBC 939凋亡抑制率。结果LY 294002作用后,5-Fu、MM C及塞莱西布对胆管癌细胞系QBC 939抑制作用明显增强,且能提高其凋亡率。结论LY 294002能有效提高化疗药物5-Fu、MM C、塞莱西布体外对QBC 939细胞抑制作用的敏感性;抑制P I3K介导的信号转导通路,可明显提高胆管癌的化疗效果。 Objective, To explore the effect of PI3K/AKT inhibitor LY294002 on increasing sensitivity of regular chemotherapeutic agent of cholangiocarcinoma. Methods, By using MTT detected the inhibition of cholangiocarcinoma when singly applied with 5-Fu,MMC,CELECOXIB contrast to associated with LY294002. At the same condition, detect the apoptosis ratio by the flow cytometer, Result: LY294002 could increase the sensitivity of the 5-Fu,MMC,CELECOXIB and increase the apoptosis ratio in vitro. Conclusion, The LY294002 can enhance significantly the sensitivity of the cyto-inhibition of the chemotherapeutic agents. Inhibiting the PI3K/AKT signaling pathway can increase the chemotherapeutic effect of cholangiocarcinoma.
