骨形态计量学观察糖皮质激素的不同剂量和不同的给药方法对大鼠松质骨的影响 被引量:2

摘要 目的通过骨形态计量学方法观察糖皮质激素的不同剂量和不同的给药方法对大鼠松质骨的影响。方法32只4月龄雌性SD大白鼠,随机公为4组:A组为正常对照组(生理盐水,按5ml·(kg·d)-1灌胃),B组为低剂量激素组(强的松2ml·(kg·d)-1,5ml/kg灌胃),C组为高剂量激素隔日组(强的松4ml·(kg·d)-1,5ml/kg的灌胃),D组为高剂量激素每日组强的松4ml.(kg·d)-1,5ml/kg的灌胃)。C组隔日给药,其余的每天一次,连续12周。实验结束时,处死全部大鼠,取胫骨中段进行不脱钙骨制片,用计算机全自动图像分析系统进行骨组织学形成计量学分析。结果与A组相比,B组各项指标无明显下降;C组的%TbAr、Tb.N、Tb.Th明显减少,Tb.Sp明显增加,说明C组大鼠骨量明显减少;D组%TbAr、Tb.N、Tb.Th、Tb.Sp均下降明显,说明D组大鼠骨量也明显丢失。C组与D组无显著性差异。结论长期使用高剂量糖皮质激素,不论是每日给药还是隔日给药均可以引起大鼠骨量减少,发生骨质疏松。长期使用低剂量糖皮质激素不引起大鼠骨质疏松。 Objective To study the effect of the adminstration method and the dose of glucocoticoid on metabolism of cancellous bone in rats. Methods Thirty-two four-month-old Sparague Dawley femal rats were randomly divided into four groups: Group A was treat with vehicle orally as control (5ml · (kg·d)^-1);Group B was administrated low-dose predispose by intragastric gavage daily (2ml ·(kg·d)^-1);Group C was administrated predispose(8 ml ·(kg·d)^-1),every other day; Group D was administrated high-dose predispose (4 ml·(kg·d)^-1) daily; Group c was administrated predispose every ofther day, others were treated once a day for 12 weeks. After 12 weeks, all rats were killed and their tibial diaphysis were processed undecalcifiled sections and measured by automatic image-analiysis system for bone histomorphometry. Results Compared with group A, no obvious changes in the index of group B; the TbAr, Tb.N, Tb. Th of group C has decreasesd significantly, but Tb. Sp increase singnificantly ; the %TbAr, Tb.N, Tb. Th, Tb. Sp of grup D has decreasesd markably, there were no significant changes in group C and group D. Conclusions The occurrence of the glucocorticoid-induced adverse reactions was closely related to the dose of gluccorticoid, no much to the administration method.
