缺铁性贫血患儿红细胞密度及红细胞超氧化物歧化酶的相应分析

摘要 采用密度梯度离心法将12例缺铁性贫血(IDA)患儿和15例正常儿童的红细胞离心分离为各1/3上、中、下三层,并采用煌焦油蓝生理盐水法。邻苯三酚自氧化抑制法分别测定其红细胞的Ret分数和E-SOD的活力,对照组和病例组的Ret上中下三层分别为0.020,0.009,0.002和0.035,0.013,0.002(P<0.01,P<0.05和P>0.05);两组的E-SOD上中下三层分别为6349,5721,5301和6850,6165,5024(P<0.05,P<0.05和P>0.05)。结果提示,上层为年轻红细胞,下层细胞逐渐衰老,IDA上层E-SOD受氧自由基影响,代偿性活力增高,IDA红细胞包含了膜LPO增加和E-SOD代偿消耗的综合病理过程。 In this study, red cells of 15 normal children and 12 IDA children were stratified into 3 layers respectively using density gradient centrifugation. Each of the layers consisted of 1/3 part of the sample. Ret fractions and E-SOD validity were measured by using normal saline brilliant cresyl blue method and O-trihydroxy benzene inhibition method respectively The top,middle and bottom layer Ret fractions of control group and IDA group are 0.020, 0.009, 0.002 and 0.035, 0.013, 0.002 respctively (P<0.01,P<0.05, P>0.05);E-SOD of the three layers from top to bottom of the control group and IDA group are 6349, 5721,5301 and 6850,6165,5024 respectively (P<0.05, P<0.05 and P>0.05). The results suggest that the top layers are young cells,and the bottom layers are aging cells.Affected by oxygen free radicals, the E-SOD validity of IDA top layer is compen-satively high.IDA red cells experienced the pathological process of membrane LPO increasing and E-SOD consumption.
