摘要 目的分析骨髓基质干细胞向神经元样细胞分化前后相关基因的表达。方法用丹参诱导骨髓基质干细胞向神经元样细胞分化,诱导后90mins,提取诱导前后的骨髓基质干细胞总RNA,RT-PCR检测ngn-1,mash-1的表达及观察其诱导前后的表达情况,免疫组化检测NSE和GFAP的表达情况。结果未经诱导的骨髓基质干细胞ngn-1,mash-1 mRNA为阴性,诱导后有表达。诱导后的细胞NSE和GFAP呈阳性反应。结论骨髓基质干细胞向神经元样的分化与ngn-1,mash-1可能有关。 Objective To investigate the gene expression of Mash-1 and Ngn-1 in differentiation of SD rat bone marrow-derived mesenchymal stem cells inducing by danshen. Methods We isolated the nucleated cells from rat bone marrow through gradient centrifugation, then induce MSCs differentiation into neurcyte-like cells by Danshen, detected the expression of specific enolase(NSE) and glial fibrillary acidic protein (GFAP) by immuncytochemistry. RT-PCR was used to detect the mRNA of Mash-1 and Ngn-1. Results The expression of Mash-1 and Ngn-1 do not detected before the inducing by danshen, and can found expression after the inducing. Both NSE and GFAP were immuncytochemistrical positive. Conclusion Mash-1 and Ngn-1 might play an important role when the MSCs differentiation into neurocyte-like cells inducing by danshen..
