摘要
目的探讨大鼠脊髓半横断(hemisected spinal cord injury,hSCI)损伤后14d硫酸软骨素蛋白多糖Versican及其受体CD44的表达变化。方法10只成年SD大鼠随机分为正常对照组和术后14d组。建立成年SD大鼠脊髓半横断(T9-T10)模型。14d后取损伤位点头尾段T9、T10节段制作冰冻切片,运用Versican、CD44抗血清进行免疫组化ABC法染色。分别观察并计数脊髓腹角和Ⅰ、Ⅱ板层内Versican、CD44的免疫反应阳性细胞数。结果Vetsican主要分布于正常大鼠脊髓腹角,CD44主要分布于正常大鼠脊髓Ⅰ、Ⅱ板层和腹角。损伤后14d腹角Versican阳性细胞数较正常组明显下降(P〈0.05),Ⅰ、Ⅱ板层和腹角的CD44阳性细胞数也较正常组明显下降(P〈0.05)。而手术组损伤侧与未损伤侧Versican和CD44表达水平均无明显差异(P〉0.05)。结论大鼠脊髓半横断损伤14d后Versican、CD44表达均明显下降,提示脊髓半横断损伤对脊髓灰质内细胞表达Versican、CD44有影响。
Objective To investigate the expression changes of Versican and its receptor CD44 in spinal cord ventral or spinal laminaⅠ、Ⅱ in 14d after hemisecfion of spinal cord injury(hSCI). Methods Ten Sprague-Dawley rats were randomly divided into normal group and hSCI group at days 14. The spinal cords were hemisected between T9 and T10. The animals were sacrificed 14 days after operation. The T9,T10 spinal segments were taken out and sectioned continuously into 25 μm transaction slices. The sections were stained with immunohistochemical ABC method, and the numbers of Versican and CD44 positive cells in spinal cord ventral horn or spinal lamina Ⅰ、Ⅱ were counted. Results Versican positive products were mainly distributed in spinal cord ventral horn in the normal group. CD44 positive products were mainly distributed in spinal ventral horn and spinal laminaⅠ、Ⅱ in the normal group. The numbers of Versican positive cells in the ventral horn in the 14d group were significantly decreased than that in the normal group( P 〈0. 05). The numbers of CD44 positive cells in the spinal lamina Ⅰ、Ⅱ and ventral horn in the 14d group were also significantly decreased than that in the normal group( P 〈0. 05). And the differentions between lesioned side and intact side of spinal cord for expression of CD44 and Versican in hSCI group were all not significant( P 〉0. 05). Conclusion The number of Versican and CD44 positive products apparently decreases in the spinal cord. It indicates hSCI may be related to the expression of Versican and CD44 for cells.
