摘要
目的研究肠型胃癌不同发展阶段基因表达谱的改变。方法应用基因芯片技术对3例肠型胃癌不同发展阶段基因表达特征进行检测分析。基因芯片包括576个cDNA克隆,其中包括506个靶克隆,51个是本课题组前期工作经消减杂交得到的克隆,另外455个克隆则根据它们在肿瘤中的作用而选取。应用Cluster和TreeView软件进行聚类分析,研究各种基因与胃癌发生发展不同阶段的关系。结果经基因芯片技术分析得到了181个差异表达基因,至少在胃癌发生发展的一个阶段其Cy5∶Cy3的比值大于2或者小于0 .5。其中48个基因上调,133个基因下调。聚类分析将差异表达基因分成了5个特征性的组,反映了不同阶段的基因表达特征。同时发现一些重要的差异表达基因, SEC23IP、LIPF、EST(BQ291520)、SLC5A1、PG、CXCR4、DICER1、SH3GL2以及IGF2R等。结论认识并发现了胃癌差异基因表达特征以及一些重要基因,对肠型胃癌发生发展及转移的进一步研究将具有重要作用。
Objective To identify gene expression patterns in distinct stages of intestinal-type gastric cancer (GC). Methods Gene expression patterns of distinct stages of intestinal-type GC samples from 3 patients were compared with cDNA microarray, which contained 576 genes. There were 506 target genes, which included 51 genes identified from our previous experiment with suppression subtractive hybridization (SSH)and other 455 genes chosen for their important roles in cancers. Hierarchical clustering was pedormed to clarify genes in association with distinct stages of GC. Results One hundred and eighty-one differentially expressed genes with average Cy5 : Cy3 ratios higher than 2.0 or lower than 0.5 in at least one stage of GC were identified by cDNA microarmy. Among them, 48 genes were up-regulated and 133 down-regulated. Hierarchical clustering analysis separated the differentially expressed genes in different stages of C,C into 5 main characteristic groups. Some important differentially expressed genes in different stages of C,C were identified, such as SEC23IP, LIPF, EST(BQ291520), SLCSA1, PG (encoding similar to pepsin A precursor), CXCR4, DICER1, 5H3GL2, and IGF2R. Conclusion The differentially expressed gene patterns and some important genes were identified, which might be useful in further study on carcinogenesis, progression and metastasis of intestinal-type GC.
