摘要
目的分析汉族人群caspase-8、-10基因的单核苷酸多态性(single nucleotide polymorphisins,SNPs)位点及其构成的单倍型,为研究caspase-8、-10基因与多基因复杂疾病的关联分析奠定基础。方法采用PCR、变性高效液相色谱技术和DNA测序技术检测caspase-10基因的第2.5外显子,caspase-8基因的第8-10外显子及其部分侧翼序列的多态性位点;分析配对位点的连锁不平衡关系,最大期望值法估算它们构成的单倍型。结果(1)caspase-10基因的第2、5外显子分别检出一个SNP位点A2823G和A12799G,其中A12799G是新发现的低信息度的SNP;caspase-8基因中检测到3个SNP位点A43466G、G51484A、G52951A,分别位于第8、9外显子和第9内含子;它们均未改变所编码蛋白的一级结构;(2)caspase-10基因中A2823G位点与caspase-8基因中3个位点间已达到连锁平衡,caspase-8基因中A43466G与G52951A、G51484A与G52951A也达到连锁平衡,连锁不平衡系数分别接近于0;只有A43466G与G51484A存在强的连锁不平衡,连锁不平衡系数接近于1;(3)caspase-10基因的A2823G位点与caspase-8基因的3个位点预计产生11种单倍型,其中A-2823/A-43466/G-51484/G-52951是主要单倍型,频率为0.381l,其次是A.2823/A-43466/G-51484/A-52951,频率为0.2536;这4个SNP位点联用,多态信息含量可达到0.7106。结论浙江地区汉族人群caspase-10、-8基因中的SNP位点至少处于3个不同的单倍型块;联合多个相邻的位点构成单倍型,可以弥补单个SNP信息度较低的不足。
Objective To investigate single nucleotide polymorphisms (SNPs) and the distribution of their haplotypes in easpase-8, -10 genes in Zhejiang Han nationality in China. Methods PCR, denaturing high-performance liquid ehromatography (DHPLC) and DNA sequencing were used to detect the SNPs in the 2nd-Sth exons of caspase-lO gene, the 8th-10th exons of easpase-8 and their flanking sequences. Expectation Maximization (EM) algorithm was used for haplotype frequencies analysis and pairwise linkage disequilibrium (LD) test. Results (1) Two SNPs, A2823G and AI2799G, were identified in easpase-10 gene, located in exon 2 and exon 5 respectively. AI2799G was newly found with low informativeness. Three SNPs were identified in easpase-8 gene; A43466G, GSI484A and G52951A were located in exon 8, exon 9 and intron 9, respectively. They do not change the primary structure of the encoded protein. (2) Linkage equilibrium was observed between A2823G in easpase-10 gene and the three sites in easpase-8 gene. A43466G and G52951A, and GSI484A and G52951A in easpase-8 gene were also in linkage equilibrium. Their coefficients of dis- equilibrium were near 0. Whereas strong linkage disequilibrium was observed between A43466G and GSI484A, because its coefficient of disequilibrium was near 1. (3) A total of 11 haplotypes were estimated within A2823G in easpase-10 gene and three sites in easpase-8 gene. A-2823/A-43466/G-51484/G-52951 was the main haplotype with a frequency of 0. 3811. A-2823/A-43466/G-51484/A-52951 was the second haplotype with a frequency of 0.2536. The polymorphism information content of their haplotypes was 0.7106. Conclusion The SNPs of easpase-8, -10 genes in Han Chinese of Zhejiang could be parsed into at least three different haplotype blocks. The polymorphism information content can be improved by using haplotype analysis of several SNPs.
