摘要
目的探讨多重聚合酶链反应(PCR)在多病原混合感染诊断中的应用价值。方法应用在 PCR 基础上改进和发展的新技术,自行设计3种特异性引物,同时扩增检出淋病奈瑟菌(NG)、解脲脲原体(UU)和沙眼衣原体(CT)DNA。结果 31例对比试验表明多重 PCR 与常规 PCR 结果符合率达97%;多重 PCR 对单一和混合感染均可检出;以多重 PCR 对209例性病患者的泌尿生殖道分泌物进行检测,检出混合感染率分别为 NG+UU 7.7%;NG+CT 3.8%;CT+UU 3.4%;NG+CT+UU 1.9%。结论多重 PCR 一次反应即可完成多次常规 PCR 检测,对于多病原混合感染的检测有简便、准确、快速、高效的特点。
Objective To rapidly detect mixed infection of Neisseria gonorrhoeae(NG),Chlamy- dia trachomatis(CT),Ureaplasma urealyticum(UU)at one test.Methods Multiplex-PCR was developed.Three pairs of primers were added in one PCR reaction system and DNA of NG,CT and UU were amplified at the same time.Results Secretion substance of urodaeum in thirty-one cases had been tested by Multiplex-PCR and conventional PCR separately,then the result showed that Multiplex-PCR were in correspondence with PCR.209 STD patients were detected by the Multiplex-PCR,suggesting that the method could determine single or mixed infection.The rates of mixed infection were NG+UU 7.7%,NG+CT 3.8%,UU+CT 3.4% and NG+UU+CT 1.9%.Conclusion As one Multiplex- PCR could finish the work that several PCR did,it was simple,accurate,rapid and effective in diagnos- ing mixed infection.We could predict that Multiplex-PCR would be a valuble method for medical labora- tory diagnosis.
