摘要 通过提纯中华蜜蜂工蜂体内的碱性磷酸酶,研究金属离子及脲对碱性磷酸酶的影响.结果表明:Na+、K+和L i+等正一价金属离子浓度为1.0-10.0 mmol.L-1时,对酶活力没有影响;Mg2+、Ca2+、Ba2+浓度为0-5.5 mmol.L-1时,对酶均有激活作用;N i2+、Mn2+、Co2+、Zn2+浓度为0-200μmol.L-1时,对酶有激活作用,而Cu2+在该浓度下对酶有抑制作用;Hg2+、Ag+及Cd2+浓度为0-150μmol.L-1时对酶有抑制作用;脲浓度低于3 mol.L-1或高于3 mol.L-1时,对碱性磷酸酶有变性失活作用. The effects of metal ions and urea on the alkaline phosphatase(AKP) from Apis eerana cerana Fabrieius were studied. The results were as follows: Na^+ , K^+ and Li^+ (1.0 -.10.0 mmol ·L^-1) had no effect on the enzyme activity; Mg^2+ , Ca^2+ , Ba^2+ (0 - 5.5 mmol·L^-1 ) could activate the enzyme; Ni^2+, Mn^2+, Co^2+ and Zn^2+ ( 0 - 200 μmol·L^-1 ) could activate the enzyme, but in this range of concentration, Cu^2+ could inhibit the enzyme; in the range of 0 - 150 μmol·L^-1, Hg^2+ , Ag^+ and Cd^2+ could inhibit the AKP; the concentration of the urea (lower than 3 mol·L^-1 or higher than 3 mol·L^-1) inactivated the enzyme.
