TGF-β1、Smad7在甲状腺癌中的表达和意义 被引量:6

摘要 目的探讨转化生长因子β1(TGF-β1)及其相关蛋白Smad7在甲状腺乳头状癌及滤泡状癌中的表达及临床病理意义。方法采用免疫组化SP法,对79例甲状腺癌及癌旁组织中TGF-β1、Smad7蛋白的表达进行检测。结果TGF-β1、Smad7蛋白表达率在癌旁组织分别为20%、16%,在甲状腺癌中为68.35%、72.15%,两者差异有统计学意义(P<0.05);不同性别、年龄、组织类型的甲状腺癌中二者表达差异无统计学意义,而在外膜侵犯、淋巴结转移、术后复发组均显著高于对应阴性组(P<0.05),并且差异有统计学意义(P<0.01)。结论TGF-β1、Smad7蛋白的表达在甲状腺癌的发生、发展中有一定作用,同时还与甲状腺癌的生物学行为密切相关,可以作为甲状腺癌诊断的辅助指标,并可能为其治疗方法的选择提供部分依据。 Objective To investigate the relationships between the biological behavior of human papillary thyroid carcinoma(PTC) and follicular thyroid carcinoma(FTC) and the protein expression of transforming growth factor β1 (TGF-β1) as well as its related downstream signal transducer- Smad7. Methods Immunohistochemical SP method was used to examine the expression of TGF - β1 and Smad7 in 79 specimens of thyroid carcinoma. Results The positive expression rate of TGF -β1, Stand7 in thyroid carcinoma were 68,35 % (54/79), 72.15 % (57/79), respectively; In pericarcinoma tissues were 20 % (6/50), 16 % (8/50), respectively, and the differentiation was significant( P 〈 0.05). In thyroid carcinoma, the groups with capsule infiltration, lymphatic metastasis and recurrence were signifi- cantly higher than the counterpart negative groups ( P 〈 0.05). There existed an obvious relationship between the expressive intensity of TGF-β1 and Smad7 ( P 〈 0.01), Conclusion The expression of TGF - β1 ,Smad7 play certain important roles in the development of thyroid carcinoma and have a significant correlation with the biological behavior of thyroid carcinoma, can be used as the reference markers in the diagnosis of thyroid carcinoma, and may provide an alternative to the treatment of thyroid carcinoma.
