摘要
利用SAS软件中二水平设计的部分重复因子实验对菌株F-1降解胆固醇的发酵培养基组成进行了初筛,发现Tween-80和酵母膏是影响降解能力的主要因子,前者为负影响,后者为正效应。然后采用中心组合设计及响应面分析确定Tween-80和酵母膏的最佳浓度分别是0.905mL/L和5.694g/L,并通过实验测得优化培养基后的降解酶活力为1.956×10-3U/mL,与预测值1.982×10-3U/mL非常接近,且比培养基组分优化前提高了22%。
A medium for enzymes of degrading cholesterol was optimized. At first, the effect of NH4NO3, Tween- 80, KH2PO4, yeast extract, MgSO4.7H2O and cholesterol on enzymes of degrading cholesterol was evaluated using a fractional factorial design, and secondly the path of steepest ascent and the response surface analysis were used to approach the optimal concentration of the medium composition. The results show Tween-80 increase the activity of enzymes, yeast extract decrese it, and optimal concentration of them is 0.905mL/L and 5.694g/L respectively. The enzyme activity of degrading cholesterol is 1.956 ×10^-3U/mL under optimized medium.
