摘要
应用MT1和MT2引物通过聚合酶链反应扩增人型和牛型结核分枝杆菌的38KDa蛋白质基因。测定了反应体系的灵敏度(0.12×102CFu/ml)和特异性。对三种不同的DNA抽提法作了比较,认为用Tween20和NP40作为裂解液与SDS法比较有显著性差异(P<0.05),且Tween20比NP40更适合。
polymerase chain reaction for identification of a 38 kilodalton protein gene of mycobacterium tuberculosis was tested by using MT1 and MT2 primers.The sensitivity(0.12× 102 cfu/ml)and specifialiy of the amplification system was detected.The results showed that the extraction of DNA with Tween 20 or NP40 had significant difference as compared with SDS(P<0.05).The extraction of DNA with Tween 20 was better than with NP40.
