淫羊藿甙对HL－60细胞增殖与分化的影响

Effects of icariin on the proliferation and differentiation of HL-60 cells

为寻找新的肿瘤细胞诱导分化剂，采用ＭＴＴ比色分析法和形态定量分析技术检测了淫羊藿甙对人急性早幼粒白血病细胞（ＨＬ－６０）增殖与分化的影响。结果表明：淫羊藿甙可以抑制ＨＬ－６０细胞的增殖，呈剂量依赖关系；选择６２．５、１２５、２５０ｍｇ·Ｌ－１进行动态观察，发现１２ｈ后即可发挥抑制作用。淫羊藿甙１００ｍｇ·Ｌ－１作用４８ｈ后ＨＬ－６０细胞ＮＢＴ还原能力明显增强（Ｐ＜０．０１），细胞核面积减小，平均光密度（ＭＯＤ）增加，积分光密度减小（ＩＯＤ）；光镜观察结果显示，淫羊藿甙作用后的ＨＬ－６０细胞发生明显变化，细胞核体积减小，细胞核呈杆状或分叶状。上述结果提示，淫羊藿甙对ＨＬ－６０细胞有诱导分化作用。

Using MTT colormetric assay and morphological quantitative analysis technique,the inhibitory and inductive effects of icariin on the human promyelocytic leukemia cell line(HL60) was observed in order to look for the new tumor cell inducer.The results showed Icariin inhibited the proliferation of HL-60 cells dosedependently;Icariin(62.5,125,250 mg·L-1)had inhibitory effects on cells after incubation 12h. Icariin(100 mg· L-1) increased the reduction of NBT(P<0.01)and MOD, reduced unclear area and IOD after incubation 48 h. The nuclear morphology of induced HL-60 cells was changed into rod or lobulated shape and the volume of nuclei reduced, suggesting icariin may induce differentiation of HL-60 cells.
