拟无枝酸菌B37与游动放线菌E92-70属间融合 被引量:1

摘要 以去甲基万古霉素产生菌B37和替考拉宁类抗生素产生菌E92-70为出发菌株,将B37原生质体热灭活,进行属间原生质体融合,融合率为1.4×10^(-6)~4.6×10^(-6),融合子50%以上抗菌活性增强。对融合子E9作了深入研究,其抗MRSA活性明显增强,纸层谱、HPLC图谱以及总DNA的SalI酶切图谱与两亲本均有差异,E9在分类上仍与E92-70同属,其产物为胞壁抑制物,作用点仍在细菌胞壁D-Ala-D-Ala上,E9很可能是B37与E92-70的重组子,其产物可能是不同于去甲基万古霉素和替可拉宁的新的糖肽类杂合抗生素。 Intergeneric protoplast fusion between Amycolatopsis sp. B37 (norvancomycin producer) and Actinoplanes sp.E92-70(teicoplanin producer)was studied. Fusants resistant to penicillin G(50Pg/ml)were found at frequency of 1.4×10-6~4. 6 × 10-6Nearly half of the antibiotic produced by fusants were improved their activity against MRSA.Fusant E9 was different from its parents on paper chromatogram. HPLC and Sal I analysis of total DNA.As its activity could be reversed by the cell wall extracts of S.aureus 209P and it was bioselectively adsorpted on Sepharose-4B-D-Ala-D-Ala, the hybrid antibiotics produced by E9 belong to glycopeptide antibiotics. Taxonomic study on E9 shows that it should be placed in the genus of Actinoplanes.
