摘要
目的:筛选卵巢老化及针刺“肾俞”干预的相关基因。方法:建立自然的更年期大鼠模型,实验动物包括青年组、更年组、针刺组,采用mRNA差异显示PCR技术,筛选卵巢中差异表达的基因,其中部分进行克隆、测序,并与NCB I相关数据库进行同源性比较。结果:获得16条差异表达明显的cDNA片段,其中已测序的6条均为未知功能基因。结论:卵巢老化及针刺“肾俞”的干预效应可能是多基因参与作用的结果,对分离到的cDNA进一步研究有助于揭示其分子机制。
Objective: To scrcen genes associated with ovary-aging and intereference of needling acupoint "Shenshu" in it. Method:The rat models for climacteric were made naturally. The experimental animals included young group,climacteric group, acupuncture group. The mRNA differential display PCR was performed to screen the differentially expressed genes of ovaries between groups. Some of them were cloned, sequenced and compared with NCBI database for homology analysis. Result :The 16 differentially expressed fragments were acquired,of which 6 fragments were sequenced and belong to function-unknown genes. Conclusion:The ovary-aging and intereference of needling acupoint "Shenyu" may result from the regulation of multi-genes. The further study to isolated cDNA fragments were helpful for uncovering their molecuar mechanism.
