摘要
目的研究含磷脂酰胆碱的玻璃酸钠制剂(PSH注射液)对兔骨关节炎(OA)的作用。方法以木瓜蛋白酶诱导兔OA模型,以2种剂量PSH注射液兔膝关节腔内注射,另设对照组。给药5周后,运用酶联免疫吸附测定法检测关节滑液中2种炎性细胞因子白细胞介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)的含量;用天青A法检测关节软骨中糖胺聚糖(GAG)总量,用放射免疫法测定其中玻璃酸(HA)的含量。结果本模型可提高滑液中IL-1β和TNF-α的含量,使关节软骨中GAG的总量和HA的含量均减少,而PSH注射液关节内注射可以降低此两种细胞因子的含量,同时使病变关节软骨中蛋白聚糖(PG)和HA的含量增加。结论PSH注射液可以降低IL-1β和TNF-α的表达,保护关节软骨,对OA治疗有一定作用。
OBJECTIVE To study the effects of sodium hyaluronate preparation containing phosphatidylcholine (PSH injections)on osteoarthritis in rabbits.METHODS The animal models of osteoarthritis were induced by papain. During the experiment, two dosages of PSH injections and controls were injected into the knee joint of the rabbits respectively. After 5 weeks of administration, the concentrations of two inflammatory cytokines, IL-1β and TNF-α, in joint synovial fluid were measured by enzyme-linked immtmosorbent assay. The contents of glycosaminoglycan(GAG) in articular cartilage, were determined by azure A colorimetry method. The contents of hyaluronic acid(HA) were measured by radioimmtmoassay. RESULTS The expression levels of IL-1β and TNF-α in joint synovial fluid were promoted in the osteoarthritis models, the quantities of total GAG and HA in articular cartilage were all declined. PSH injections significantly inhibited the expression levels of these cytokines, and increased the quantities of proteoglycan (PG) and HA in pathological articular cartilage. CONCLUSION PSH injec- tions has remedy effects on osteoarthritis rabbits by inhibiting the expression levels of IL-1β and TNF-α and protecting the articular cartilage.
