非小细胞肺癌内皮素对血管内皮生长因子的影响及与病理特征的关系 被引量:2

摘要 目的研究内皮素(Endothelin-1,ET-1)与血管内皮生长因子(Vascular endothelial growth factor,VEGF)在肺癌的阳性表达、影响肺癌生长转移的作用机制及其相互作用关系。方法应用免疫组化SABC染色法检测40例原发性支气管肺癌标本中ET-1、VEGF的阳性表达、微血管密度(Microvessel density,MVD)计数,分别评价ET-1和VEGF与非小细胞肺癌临床病理参数之间的关系、ET-1和VEGF的关系;采用图象分析技术测定ET-1含量。结果ET-1和VEGF在肺癌组织中呈高阳性表达,二者阳性表达者的MVD值均明显高于VEGF阴性者,差异有统计学意义。ET-1在非小细胞肺癌组织中呈现阳性高表达,其含量及阳性表达与肿瘤分化程度、大小和淋巴结转移密切相关。分化程度越低,肿瘤直径越大、已有淋巴结转移,则ET-1含量和表达越高。在非小细胞肺癌VEGF阳性率与肿瘤大小,淋巴结转移与否密切相关。ET-1表达阳性区域,VEGF表达也呈阳性,在两者皆表达阳性的癌组织中MVD值明显高于单一阳性表达的肺癌组织,差异具有统计学意义。结论(1)ET-1在促进非小细胞肺癌血管生成的过程中发挥重要作用,可作为评估非小细胞肺癌恶性程度、淋巴结转移潜能和预后的重要监测指标之一;(2)ET-1的表达与VEGF有关,可能对VEGF具有上调作用,二者相互协同,共同调节和诱导肺癌微血管的生成。 Objective To study positive expression of endothelin - 1 ( ET- 1), relationship between the ET - 1 and vascular endothelial growth factor (VEGF), and discuss the role of ET- 1 in neovascularzation of lung cancer. Methods ET- 1 and VEGF positive expressions were detected by immuno - histochemistry and image analysis. Results ET - 1 positive expressions were mainly located in the cytoplasm of lung cancer. The positive rate of ET- 1 in non - small cell hing cancer was significantly higher than that in small - cell lung cancer. The larger the tumor, the lower the differentiating grade, and the more extensive the metastasis of non- small cell lung cancer was, the higherasas ET - 1 positive expression. Microvessel demity was significantly higher in the tissue specimens of both ET- 1 and VEGF positive expressions. Condusion ET- 1 plays an important rolle in the process of growth and metastasis of non - small cell lung cancer. ET- 1 may lead to VEGF overexpression in lung carcinoma, which contributes to neovascalarization of non - small cell lung cancer as an angingenic regulator.
