精神分裂症患者伏隔核的DARPP-32及其磷酸化水平 被引量:2

DARPP-32 Protein and Phosphorylation of DARPP-32 in the Nucleus Accumbens of Patients with Schizhophrenia

摘要 目的探讨DARPP32蛋白及其苏氨酸磷酸化水平和精神分裂症症状的关系。方法取17例精神分裂症患者(I型7例,Ⅱ型10例)和9例对照组的脑组织,蛋白质免疫印迹法测定伏隔核中DARPP32、34PDARPP32和75PDARPP32的水平。结果在脑的伏隔核,对照组、I型组、Ⅱ型组的DARPP32分别为(100.0±7.3)、(70.8±4.7)和(96.2±5.8),(单因素方差分析P<0.05);34PDARPP32分别为(100.0±8.7)、(66.3±7.0)和(102.0±9.2),(单因素方差分析,P<0.05);而75PDARPP32分别为(100.0±19.9)、(79.8±7.0)和(118.1±22.0),(单因素方差分析,P>0.05)。结论DARPP32及其在34苏氨酸磷酸化程度的减少和精神分裂症的阳性症状有关。 Objective To explore the association between DARPP-32(dopamine and cAMP-regulated phosphoprotein,32 kD) threonine phosphorylation and the symptoms in schizophrenia. Methods DARPP-32, 34-phosphothreonine-DARPP-32, and 75-phosphothreonine-DARPP-32 were immunoquantilied in the nucleus accumbens of postmortem brains obtained from 17 patients with schizophrenia( Ⅰ type 7 patients, Ⅱ type 10 patients) and 9 controls. Results In the Ⅰ type schizophrenia, DARPP-32[-controls (100. 0+±7.3), Ⅰ type(70.8±4.7), Ⅱ type(96.2±5.8), P〈0.05] or 34-phosphothreonine-DARPP-32 [controls(100.0±8.7), Ⅰ type(66.3±7.0), Ⅱ type(102.0±9.2), P〈0.051 were significantly decreased in Ⅰ type schizophrenia than in controls or in Ⅱ type schizophrenia. Conclusion Decreased of DARPP-32 phosphorylation at the 34-threonine in the nucleus accumbens may be connected positive symptom of Ⅰ type schizophrenia.
