摘要
目的用荧光光度法测定芦荟康胶囊中的芦荟大黄素和芦荟苷的含量。方法在pH为5.76的HAcNaAc缓冲液中和十二烷基磺酸钠(SDS)表面活性剂存在下测定芦荟大黄素的λex和λem。在Na2B4O7NaOH介质中测定芦荟苷的λex和λem。结果芦荟大黄素和芦荟苷浓度分别在1.0×106~2.5×105mol/L和1.0×106~2.0×105mol/L,与其荧光强度呈线性关系,检测限分别为2×108mol/L和1×108mol/L。芦荟大黄素的λex和λem分别为460,550nm,芦荟苷在此体系无荧光;芦荟苷的λex和λem分别为387,533nm,芦荟大黄素在此体系无荧光。结论该法灵敏度高,选择性和重现性好,可直接测定芦荟康胶囊中芦荟大黄素和芦荟苷的含量。
Objective The flurimetric determination of aloe-emodin and aloin in aloe capsules was described. Methods In the presence of pH 5.6 HAc-NaAc buffer and SDS surfactant, excitation and emission maximal of aloe-emodin were at 460 nm and 550 nm(λex/λem=460/550 nm). Aloin in this system do not emit fluorescence. In the presence of pH 5.6-9. 2 NaOH-Na2B4O7 buffer, aloin reacts with borax, the system fluorescence intensity was 10 times that of origin, λes/λem=387/583 nm. Results The linear range of aloe-emodin was 1.0 × 10^-6-2. 5 × 10^-5 mol/L in 460/550 nm and aloin was 1.0 × 10^-6 -2.0 × 10^-5 mol/L in 387/583 nm. Its detection limit was 2 × 10^-8 mol/L in aloe-emodin and 1 × 10^-8 mol/L in aloin. Conclusion This method is sensitive, repetitive and simple. It can be applied to the determination of aloe-emodin and aloin in aloe capsules.
