摘要
将编号为01,02,03,04,05的5个伊氏锥虫克隆群体以107条/只的剂量经耳静脉分别接种至1只新西兰大白兔体内,免疫荧光试验检测抗体滴度达到最高后,开始耳静脉采血提取抗血清。之后每隔2天提取一次抗血清,每只共提取10次,5只共50份血清等量(各占1/50)混合,称为抗活体锥虫血清“抗体库B”;此外取BeTat1·1,BeTat1·2……BeTat1·19,BeTat1·20共计20个抗原变异体以107条/只的剂量经耳静脉分别接种至1只新西兰大白兔体内,免疫荧光试验检测抗体滴度达到最高后,一次心脏采血,分离血清,20只共20份血清等量(各占1/20)混合,称为抗活体锥虫血清“抗体库A”;取编号为01的伊氏锥虫克隆群体以100条/只的剂量腹腔注射分别接种小鼠和大鼠,镜检见虫后使用抗活体锥虫血清“抗体库B”和抗活体锥虫血清“抗体库A”分别进行治疗。结果显示“抗体库B”治疗组的小鼠保护率为93·75%,大鼠的保护率达到了100%,“抗体库A”的治疗效果则不明显。
New Zealand rabbits were inoculated with BeTat 1.1, BeTat 1.2 BeTat 1. 19 or BeTat 1.20 VATs of Trypanosome evansi respectively. Twenty sera were obtained from every rabbit ear vein blood when the antibody titer reached the peak detected by IFA (immunofluorescence assay). Rabbit anti-T, evansi antibody "bank A" was consisted of equal parts of the 20 sera. At the same time, cloned Trypanosome evansi population numbered as 01, 02, 03, 04 or 05 was also inoculated into into New Zealand rabbits respectively. Ten sera were harvested from each rabbit with the same method at 2-day interval since the antibody titer reached the peak. and then, total 50 parts of sera were obtained and equal parts of all the 50 sera were mixed as Rabbit antiTrypanosome evansi "antibody bank B'. Rats and mice were challenged via the abdominal cavity with the cloned T. evansi population coded 01 at a dosage of 100 parasites per animal, then treated with Bank A or Bank B respectively when trypanosomes were detectable. Results showed that 93.75% mice and 100% rats treated with Bank B survived, while those treated with Bank A failed to survive.
