铁蛋白对卵巢癌细胞增殖组分及超微结构的影响 被引量:5

摘要 应用免疫组织化学法测定增殖细胞核抗原(PCNA)及电镜观察,了解H及L铁蛋白分别对人上皮性卵巢癌细胞株ao及3ao增殖组分及超微结构的影响。结果:发现H及L铁蛋白均使ao及3ao细胞增殖组分减少(P<0.05)。并发现:H及L铁蛋白可使ao及3ao细胞线粒体及糖原颗粒减少,溶酶体及脂滴增加。H铁蛋白还可使细胞内质网排列紊乱,部分扩张,核糖体脱失并使细胞核固缩;核膜皱缩及异染色质增加,边聚。提示:铁蛋白,特别是H铁蛋白,可使卵巢癌细胞增殖降低并使其能量代谢及蛋白质合成发生障碍,细胞自身清除作用增强,使核趋向不活跃。因此,可能铁蛋白作为一种抑素样物质,对卵巢癌细胞的增殖起抑制作用。 bjective:To detect the effects of the ferritin subunit H and L on ovarian cancer cells(ao and 3ao)by using the methods of immunohistochemical staining and electron micro-scope,Methods:The ao and 3ao cells were grown in a 24 well tissue cuIture plates(5×10 ̄5 cell/well)and exposed to ferritin subunit H and L(100ng/ml) for 72 hours,then the cell suspensions were centrifuged and the supernatant decanted,the cells were preserved for im- munohistochemical staining and microscopic observation. Result:The result showed that the ferritin could decrease the growth fraction of ovarian cancer cells(ao and 3ao)and their mi-tochondria and glycogen,and increase the lysosome, Besides these,the ferritin subunit H not only caused the rough endoplasmic reticulum expansion and irregular arrangement,but also increased heterochromatin. Conclusion:Ferritin,especially the subunit H may be as a chalone,suppresses the growth of ovarian cancer cell and block their engeric metabolism andthe synthesis of protein.
