摘要
为有效地监测肉类产品中的药物残留,建立了测定猪肉中安定残留量的反相高效液相色谱法。利用Al2O3层析柱对安定提取液进行净化,高效液相色谱仪测定参数:C18柱(2mm×150mm,3μm);流动相:甲醇+水+25mmoll乙酸铵溶液(64.8+35.0+0.2);流速0.2mlmin;光电二极管阵列检测器,波长254nm。实验结果表明,提取液中各组分分离良好,安定浓度在100ngml~40μgml范围内与峰面积呈线性相关,r=0.99719;平均回收率80.8%;RSD<5%。该方法快速、简单、准确,适用于肉类产品中安定残留量的检测。
The extraction sample was purified by Aluminum oxide column for chromatography and analyzed by RP-HPLC. The HPLC analyzing parameters included ODS column (2 mm × 150 mm i. d. , 3μm), the mobile phase was methanol + water + 25 mmol/L ammonium acetate (64.8 + 35.0 + 0.2), at a flow rate of 0.2 ml/miu, the DAD detection wavelength was 254 nm. The results indicated that the separation of components in the sample was good, the concentration of Diazepam within the range from 100 ng/ml to 40μg/ml was linearly correlated with the peak area, r = 0.997 19, the average recovery was 80.8% and the RSD 〈5%.
