以苯丙酮为起始原料,经溴化、Ullmann缩合、重排、水解、合成非诺洛芬,再成钙盐,总收率41.5%。亚探讨了Lewis酸对重排的影响。
A mild and convenient preparation of fenoprofen calcium is disclosed. Propiophenone via bromination, Ullmann condensation, 1,2-aryl rearrangement hydrolysis and salt formation produced penoprofen calcium in 41.5% overall yield. It was found that, the quantity of I2 was reduced and reaction time was shortened notably by adding Lewis acid in the rearrangement step.
