摘要
采用4种动物实验性心律失常模型,观察了二乙酰关附甲素(关附乙素,DGFA)的抗心律失常作用。结果表明,DGFA能显著对抗乌头碱诱发的大鼠室性早搏、室性心动过速和室性纤颤;明显对抗氯化钙诱发的大鼠室颤和氯仿诱发的小鼠室颤;明显对抗大鼠结扎左冠状动脉所致心律失常,减少室性早搏的发生次数,延迟首次室性早搏的出现时间,降低室生心动过速的发生率。其作用机制可能主要与其阻滞心肌细胞的Na+、Ca2+通道有关。
Guanfubase A is isolated from Aconitum coreanum (Lev1.) Raipaics.We studied the antiarrhythmic effects of diacetylguanfubase A (guanfubase B, DGFA).DGFA (iv) could markedly antagonize aconitine-induced ventricular premature beat (VP), ventricular tachycardia (VT) and ventricular fibrillation (VF) in rats. DGFA could remarkably antagonize CaCl_2-induced VF in rats and chloroform-induced VF in mice, antagonize arrhythmias induced by coronary ligation in rats, reduce the frequency of VP, delay the occurrence of VT.The therapeutic index of DGFA is 17.The study suggested that it may have depression on influx of Na+ and Ca2+.
