摘要
目的探讨选择性环氧化酶2(COX2)抑制剂尼美舒利对肝癌细胞SMMC7721生长的影响及其作用机制。方法MTT法检测对照组和实验组(尼美舒利25、50、100、200、400μmol/L)作用24、48、72h后SMMC7721细胞增殖的抑制率,RTPCR、ELISA及RIA检测药物作用48h后SMMC7721细胞分泌的血管内皮生长因子(VEGF)、前列腺素E2(PGE2)的变化。结果尼美舒利对肝癌细胞SMMC7721的增殖有抑制作用,且呈剂量和时间依赖性;能降低肝癌细胞SMMC7721分泌的VEGF、PGE2水平。结论选择性环氧化酶2抑制剂尼美舒利可以抑制肝癌细胞的生长和降低VEGF水平,并能降低COX2的下游产物PGE2水平。
Objective To study the effects of nimesuli, a selective cyclooxygenase inhibitor, on the growth of human hepatocelluar carcinoma cell line SMMC-7721. Methods SMMC-7721 cells were treated with nimesuli (25 μmol/ L,50 μmol/L,100 μmol/L,200 μmol/L, 400 μmol/L) for 24 h,48 h and 72 h,MTT reduction assay was used to evaluate the inhibitory rate;Mter 48 h treatment of nimesuli in SMMC-7721,the expression of VEGF mRNA and protein of SMMC-7721 cells were examined by RT-PCR and ELISA, and the expression of PGE2 was analysed by RIA,respectively. Results The inhibitory rate of nimesuli on SMMC-7721 cells proliferation increased as incubated time and nimesuli concentration increasing ;VEGF and PGE2 expression in SMMC-7721 cells were inhibited in a concentration dependent pattern by nimesuli. Conclusion The selective cyclooxygenase inhibitor, nimesuli, can inhibit the growth of SMMC-7721 cells and decrease the expression of VEGF,while it also reduce the levels of COX- 2-derived PGE2.
