摘要
目的探讨应用生物芯片扫描仪定量检测肿瘤组织芯片荧光信号的可能性。方法将49例乳腺癌及29例癌旁组织制成组织芯片,分别采用免疫荧光组织化学技术和免疫酶组织化学技术对组织金属蛋白酶抑制剂1(TIMP1)进行染色,并应用生物芯片扫描仪和显微图像分析系统分别对上述染色结果进行定量分析,同时应用半定量积分法判定染色结果。结果组织芯片中49例癌组织TIMP1免疫荧光染色用生物芯片扫描仪定量分析的结果与免疫酶组织化学染色图像定量分析结果呈正相关(P<0.01);两种方法半定量结果检测效果也一致。结论生物芯片扫描仪可用于肿瘤组织芯片荧光信号的相对定量分析。
Objective To investigate the possibility of quantification for a target protein showed by immunostaining on tissue microarray (TMA) by use of a microarray scanner. Methods A TMA contained 13 ×6 tissue cores was constructed with tissues of 49 cases of breast cancer and 29 normal tissues. These tissues were detected for the expression of tissue inhibitor of metalloproteinase-1 ( TIMP-1 ) using immunofluorescence histochemistry (F-IHC) and immunoenzymatic histochemistry (E-IHC). The results were quantificationally analyzed by a microarray scanner and a micro-image analysis system respectively. Semi-quantitative analyses for the two different staining results were performed simultaneously. Results TIMP-1 content showed by F-IHC and quantitatively measured by microarray scanner was positively related to that showed by E-IHC and quantitatively measured by micro-image analysis ( P 〈 0. 01 ), which was positively associated with the semi-quantitative result as well. Conclusion Microarray scanners might be an useful tool for quantificational evaluation of protein contents detected on tumor TMAs.
