摘要
目的 观察不同年龄段正常SD大鼠眼前段组织中MMP-2、TIMP-1和TIMP-2的表达,探讨正常生理状态下MMP及TIMP在小梁网房水流出及葡萄膜巩膜房水流出通道中的作用。方法 应用酶免疫组化技术,检测不同年龄段正常SD大鼠眼前段组织中MMP-2、TIMP-1和TIMP-2的定位。阳性结果应用图像分析系统进行定量分析。结果 免疫组化结果显示MMP-2、TImP-1及TIMP-2分布于SD大鼠眼前段的虹膜、睫状体及小梁网。MMP-2在眼组织各部位的阳性反应强于TIMP-1及TIMP-2。随着年龄增长.MMP-2、TIMP-1及TIMP-2表达量逐渐增加,在出生后2~3月(成年期)达最高值.以后(中老年期)出现下降。结论 MMP-2、TIMP-1和TIMP-2广泛分布于不同年龄段SD大鼠的小梁网房水流出及葡萄膜巩膜房水流出通道,且随着年龄变化而改变,对维持正常厉水流出具有重要作用。
Objective To investigate the expression of metrix metalloproteinase-2( MMP-2 ) and tissue inhibitor of metalloproteinase-1,2 ( TIMP-1,2 ) in anterior segment tissues of normal rat eyes in different ages and to determine the roles of MMP and TIMP in trabecular outflow and uveoscleral outflow in normal eyes. Methods Immunohistochemistry was used to detect the presence of MMP-2 and TIMP-1,2 in anterior segment tissues of normal rat eyes in different ages. The immunohistochemical staining was further analyzed and the intensity of reaction products were detected with an image-processing system. Results MMP-2 and TIMP-1,2 were expressed in the iris,ciliary body, trabecular meshwork. The staining of MMP-2 was stronger than TIMP-1,2. The expression of MMP-2 and TIMP-1, 2 increased from infant to adult (2- 3 months), then decreased. Conclusion MMP-2 and TIMP-1,2 were prominently identified in regions of the anterior segment of normal rat eyes associated with the trabecular outflow and uveoscleral outflow pathway, as well changed with ages. These data support the hypothesis that the balance between MMPs and TIMPs may be important in maintaining homeostasis in the outflow pathway.
