摘要
通过将免疫小鼠的脾细胞与HGPRT缺陷的NS-1骨髓瘤细胞进行细胞融合,并经HAT培养液对融合的杂交瘤细胞进行选择培养,再经二次限界稀释法对此杂交瘤细胞进行克隆化。采用无血清细胞培养技术,得到较高浓度而且较纯化的抗家兔晶体上皮细胞膜单克隆抗体。为进一步研究晶体上皮细胞的特性,以及利用免疫学方法抑制白内障术后晶体上皮细胞增殖奠定了基础。
The spleen cells from immunized mice were fused with HGPRT deficiency mouse myeloma cells (NS 1 ). Selection of NS- 1: spleen cell hybrids was accomplished by culturing the fusion mixture in HAT medi um. The positive hybridomas were cloned by limiting dilution method. The purified monoclonal antibodies were got by using serum-free medium cell culture. This method would be helpful to study the specialities of lens epithelium and also useful to make resarches on the prohibitive effects of monoclonal antibody on proli feration of lens epithelium.
