PD鼠纹状体内NMDA受体密度及黑质内NMDA受体mRNA含量的观察 被引量:2

An observation of density of N-methyl-D-aspartate receptors in striatum and mRNA level of N-methyl-D-aspartate receptors in substantia nigra of rats with Parkinson's disease

摘要 采用核酸斑点杂交定量法和放射配体饱和分析法,分别测定了6-羟基多巴胺(6-OHDA)损毁一侧黑质纹状体通路后同侧黑质内N-甲基-D-门冬氨酸(NMDA)受体mRNA含量及纹状体内NMDA的受体密度,研究发现该通路损毁两周后,NMDA受体密度减少,而NMDA受体mRNA含量无明显改变;损毁3个月后,两者均明显减少。研究结果提示NMDA受体密度于早期减少,反映了该通路多巴胺能神经纤维的降解,损毁后期两者减少,说明NMDA受体位于该通路神经纤维的突触前膜。 After the unilateral striatum passway in substantia nigra was destroyed with 6-hydroxyDopamine,the density of N-methyl-D-aspartate (NMDA)receptors susceptible to glutamlnic acid in striatum and the mRNA level of NMDA receptors in substantia nigra of rats were measured respectively with quantitative nucleic acid spot hybridization and radioligand binding assay.It was found that:(1)Two weeks after the destruction,the density of NMDA receptors in the ipsilateral striatum was decreased whereas the mRNA level of NMDA receptors in the ipsilateral substantia nigra remained the same.(2)Three months after the destruction,both of them were significantly decreased. The fact that the density of NMDA receptors in ipsilateral striatum began to decrease at the early stage after the destruction suggests that the dopaminergic fibers in the passway degenerated. While both were decreased at the late stage after destruction suggests that NMDA receptors are located on the synaptic anterior membrane of the neural fibers in the passway.
