摘要
目的利用蛋白质组学的研究方法建立分辨率高和重复性好的肝大部切除术后小鼠血浆的双向凝胶电泳图谱,描述肝大部切除后早期血浆蛋白质组变化的概况。方法以肝大部切除术方法制备小鼠模型,对12 h后切肝组及对照组小鼠血浆样品分别进行双向凝胶电泳,每组设平行胶3块,凝胶经银染显色后,Imaging Master 2D图像分析软件进行比较分析,识别差异表达的蛋白质。结果与对照组比较,切肝后12 h小鼠血浆蛋白中新增蛋白点37个,缺失15个,浓度上调蛋白点4个,下调7个,总的差异表达蛋白点数为63个。结论本研究建立了分辨率高且重复性较好的肝再生小鼠的血浆蛋白质组学双向凝胶电泳图谱,发现了一批与对照组相比差异表达的蛋白质,为进一步分析鉴定与肝脏再生启动相关的关键蛋白,探寻肝脏再生的机制奠定了基础。
Objective To investigate the early proteinomics of plasma of mice with liver injury, two-dimensional dectrophoresis profiles of plasma from the mice undergoing partial hepatectomy (PH) were established. Methods Kunming mice underwent either sham operation or PH. The phsma proteins from both groups were collected 12 h after operation, and separated by two-dimensional electrophoresis gds, respectively. After silver staining, the differential expression of proteins was analyzed by using Imaging Master 2D analysis software. Results Compared with the sham group, 63 differences in the protein profiles were observed in PH group. Of them, 37 protein dots were newly found, 15 dots disappeared, while 4 dots were up-expressed and 7 proteins down-expressed. Conclusion Two-dimensional electrophoresis profiles of plasma proteins with high resolutions and repetitiveness were established from mouse model. Some differential protein expressions between the two groups were observed, which was beneficial to further studies of the specific biomarkers of liver regeneration.
