双自杀基因重组腺病毒对肺癌细胞的杀伤作用 被引量:3

摘要 目的构建含CD/UPRT双自杀基因的重组腺病毒Ad-CD/UPRT,观察其对人肺腺癌细胞的杀伤作用。方法用0-100感染复数的重组腺病毒转染肺腺癌A549细胞后加入(0-1 000)mg/L的5-氟胞嘧啶(5-FC),MTT法检测杀伤效率;将转基因细胞与未转基因细胞按不同比例混合,观察旁观者效应。结果 Ad-CD/UPRT转染A549后对细胞生长有显著的抑制作用,细胞杀伤效率随腺病毒滴度及5-FC浓度的增加而增强;转基因细胞占混合细胞10%以上,即可观察到明显的旁观者效应。结论 Ad-CD/UPRT联合5-FC对肺腺癌A549细胞有显著杀伤作用。 Objective To construct a recombinant adenovirus vector containing CD/UPRT double suicide gene and to evaluate the specific killing effects on pulmonary carcinoma in vitro. Methods A549 cells were infected with 0, 1, 10,100 MOI of Ad-CD/UPRT and 0, 10,100, 1 000 mg/L concentrations of 5-FC. After 4 days of incubation, MTT assays were performed to measure the cell viability. Resuits The cell killing effects depended on Ad-CD/UPRT dose and the 5-FC concentration. Mixed cellular assay revealed that CD/UPRT-positive cells had a bystander effects on CD/UPRT-negative cells. Conclusion Ad-CD/UPRT and 5-FC system can effectively suppress the pulmonary carcinoma cell growth.
