摘要 目的:制备白细胞介素-2(IL-2)前体脂质体并考察其稳定性。方法:采用冷冻真空干燥法制备IL-2前体脂质体,采用Sephadex^(TM) G-50柱分离脂质体,MTT比色法测定活性,激光粒度仪测定粒径。结果:制备的IL-2前体脂质体为白色的非定型粉末,粒径分布在0.1~1.0μm,包封率为95.5%,活性5.0×10~6IU·g^(-1)。结论:本方法制备的前体脂质体制剂有较高的包封率,贮存稳定,粒度分布良好。 Objective :To prepare and characterize the interleukin-2 pro-liposome. Methods :The IL-2 pro-liposome was prepared by the vacuum freeze-drying system and then purified by the Sephadex^TM G-50 column. The interleukin-2 activity was measured using the MTT colorimeter, and the particle diameter was determined by laser diffraction systems ( HORIBA LA-920). Results :The IL-2 pro-liposome was non-finite white powder with the particle diameter of 0.10 - 1μm and the activity of five millions unit/g. The inclusion rate of IL-2 pro-liposome was 95%. Conclusion:The Interleukin-2 pro-liposome with higher inclusion and stability was achieved.
