摘要
应用阿霉素所致大鼠肾病综合征(NS)模型与腹主动脉-下腔静脉造瘘所致大鼠充血性心力衰竭(CHF)模型,研究了精氨酸血管加压素(AVP)系统在其中的变化以及黄芪的保护作用。斑点杂交显示下丘脑AVPmRNA表达在NS增加,在CHF降低。逆转录-聚合酶链反应表明肾脏皮质AVPV1α受体mRNA表达在NS增加,在CHF降低。黄芪对NS中AVP系统的高表达与CHF中AVP系统的低表达均有改善作用。上述变化均不是通过渗透性调节机制,心房利钠肽(ANP)在其中亦不是起主要作用,对此进行了讨论。
The changes of arginine vasopressin(AVP)system in adriamycin-induced nephrotic syndrome(NS)and aortacaval fistula-caused high-output congestive heart failure(CHF)and the protective effects of Chineseherb astragalus membraneceus(AM)were investigated.Dot blot analysis showed the significant increment of hy-pothalamic AVP mRNA expression in NS while decrement in CHF. RT-PCR demonstrated that the mRNA expres-sion of AVP V1a receptor in renal cortex was markedly enhanced in NS, while in CHF it was reduced,AM couldnotably ameliorated both the higher expression of AVP system in NS and the lower expression of that in CHF. Allthese changes were regulated through non-osmotic mechanisms,also atrial natriuretic peptide did not play the im-portant role,which was fully discussed in the article.
