高效液相色谱法测定不同产地柴胡的皂苷a含量

Determination of saikosaponin a in Bupleurum by HPLC

目的：建立柴胡皂苷a的含量测定方法，并测定41种不同产地柴胡中的皂苷a含量。方法：采用高效液相色谱法（HPLC），伊利特C18柱，以乙腈水（40：60）为流动相，流速：1．0mL·min^-1，检测波长：210nm。结果：柴胡皂苷a在1．92～19．20pg范围内呈良好的线性关系，r=0．9994。平均回收率为％．74％。RSD为1．6％。结论：不同产地柴胡中的皂苷a含量差异较大。

OBJECTIVE To establish a method to determine of saikosaponin a in Bupleurum from 41 samples in Gansu. METHODS Yilite-C18 column was used,the mobile phase consisted of acetonitrilc-water （40：60）,the flow rate was 1. 0 mL· min^-1 and detection wavelength was set at 210 nm. RESULTS The linear range of saikosaponin a was 1.92 - 19. 20 μg（r = 0. 999 4）, the average recovery and RSD value of saikosaponin a were 96.74% and 1.6%. CONCLUSION Contents of saikosaponin a from various origin are different significantly.
