缺氧对鼠胚大脑神经元c-Fos表达的影响及当归保护作用 被引量:6

摘要 目的:探讨宫内缺氧对胚胎大鼠大脑神经元内c-Fos表达的影响及当归注射液是否对缺氧鼠胚大脑神经元有保护作用。方法:将孕15d孕鼠采用低张性缺氧模型致鼠胚宫内缺氧,鼠胚大脑组织经c-Fos和神经元特异性标记物-神经元特异烯醇化酶(NSE)免疫组化双标染色后,进行图像分析。结果:三组鼠胚大脑中神经元特异烯醇化酶阳性细胞数量差异没有显著性;而缺氧组中c-Fos/NSE双染阳性细胞比对照组增多(P<0.05);当归组双染阳性细胞比缺氧组减少(P<0.05)。结论:缺氧可刺激鼠胚大脑神经元内c-Fos的表达。当归注射液对缺氧鼠胚大脑神经元可能有保护作用。 Objective:To study the effect of hypoxia in uterus on the expression of c-Fos in neurons of cerebrum from rats'embryos and whether Angelica sinensis injection has the protective role to neurons in hypoxia. Methods: The 15d-conceiving female rats were in hypotonic hypoxia in uterus. The cerebrum tissue of rats' embryos was double stained with c-Fos and neuro-specific enolase (NSE),and then the results of double staining were analyzed by image processing system. Results: The expression of NSE in 3 groups had no discrepancy in statistics. But the double staining postive cells with c-Fos/NSE in hypoxia group were more than that of control group(P〈0. 05);and in Angelica group, they were less than that of hypoxia group(P〈0. 05). Conclusion :①Hypoxia could stimulate the expression of c-Fos in neurons of cerebrum from rats' embryos. ②Angelica sinensis injection could play a protective role to neurons in cerebrum of embryonic rats in hypoxia.
