γ射线照射对单采血小板保存中转化生长因子-β1 被引量:1

Effect of Prestorage Gamma Ray Radiation on the Transforming Growth Factor-β1 Levels in Stored Apheresis Platelets

摘要 目的研究γ射线照射对单采血小板保存过程中转化生长因子-β1(TGF-β1)含量的影响。方法应用COBESpectraTM细胞分离机单采血小板,以27Gy的γ射线照射,于标准条件下保存5d,同时设自身对照,间隔取样,采用ELISA方法检测TGF-β1含量。结果对照组单采血小板在保存1d后,TGF-β1含量与保存前比较升高有显著性(P<0.05),照射组保存3d后,TGF-β1含量与保存前比较差异无显著性(P>0.05)。保存5d后,TGF-β1与保存前比较升高有显著性(P<0.05)。在保存中的各时间点,其含量均显著低于对照组(P<0.05)。结论27Gyγ射线照射能抑制单采血小板保存过程中TGF-β1含量的升高,可以有效地预防单采血小板输注后引起的免疫抑制。 Objective To study the effect of prestorage gamma radiation on the transforming growth faetor-β1 (TGF-β1) levels in storage apheresis platelets. Methods Apheresis platelets were prepared by the COBE spectra^TM cell separator,then had been irradiated by gamma ray at the dosage of 27 Gy stored at standard condition for 5 days. TGF-β1 levels were measured by ELISA at various storage times. Results The contents of TGF-β1 in apheresis platelets of control group after 1 days storage were significantly different from those of prestorage (P〈0.05). TGF-β1 in apheresis platelets of irradiated group after 3 days compared to those of prestorage showed no significant change (P〉0. 05), but after 5 days storage,the contents of TGF-β1 were much higher than those of prestorage (P〈0.05),and the contents of TGF-β1 were much lower than those of control group during storage(P〈0.05). Conclusion Gamma radiation at 27 Gy is effective in preventing the generation of TGF-β1 in the storage of apheresis plateletso And it could prevent the immunosuppressive effect of platlet transfusion.
