摘要
目的从21味药中筛选对α-葡萄糖苷酶具有活性的中药。方法选用对α-葡萄糖苷酶活性抑制程度较高的中药枸杞作为研究对象。采用烘干、回流脱脂、水提、乙醇沉淀和DEAE纤维素柱层析等方法进行枸杞多糖的提取,并对其活性以及在不同条件下对α-葡萄糖苷酶的影响进行研究。结果0.4 mg的枸杞多糖对α-葡萄糖苷酶的抑制程度达52%,2.0 mg的抑制率高达88%。结论枸杞多糖对α-葡萄糖苷酶具有较强的抑制作用,抑制机理为非竞争性抑制。
OBJECTIVE To screen Lycium barbarum from 21 Chinese traditional medicines to inhibit α- glueosidase.METHODS Lyciurn barbarum polysaccharides(LBP) were extracted by drying , circumfluence degrease, distillation with distilled water, ethanol deposition and chromatography of diethylaminoethanol(DEAE) ion- exchange cellulose. LBP effects on α-glucosidase activity under different conditions were studied. RESULTS Inhibition ratio of α-glucosidase was 52 % and 88 % at a dose of 0.4 mg LBP and 2 mg LBP , respectively. CONCLUSION LBP is a stronger α-glueosidase inhibitor. Its inhibitive mechanism is noncompetition inhibition.
